    Evaluation des techniques biophysiques pour l'analyse du repliement des protéines. Outils : Fluorescence, Nano-DSF, Nano-DSC, Dichroïsme Circulaire, Spectroscopie de masse, Mass-HDX.Certains procédés de production de protéines recombinantes à visées thérapeutiques utilisent la bactérie Escherichia coli comme outil de production. Celle-ci se réalise via la formation de corps d'inclusion de la protéine au sein de la bactérie. Dans cet état, la protéine n'a pas acquis sa structure tridimensionnelle finale, dite "native", permettant son activité biologique et donc son effet thérapeutique recherché. Une étape de repliement in vitro est alors nécessaire en amont du procédé de purification afin que la protéine adopte sa conformation native. Cependant, il n'existe à ce jour aucune technique de routine permettant de contrôler le bon déroulement de cette étape. L'objectif de ce projet a donc été d'étudier 5 techniques de caractérisation de la structure des protéines sur leurs applications possible dans le contrôle de l'état de repliement des protéines au cours du procédé de fabrication d'une protéine thérapeutique.

    Innobiochips, a company created in 2008, is specialized in the conception of multiplex immunodiagnostic tools: biochips. This technology relies on the absorption of biomolecules (peptides, protein or cells) on a treated surface according to a patent-protected process. The company has developed and commercialized in 2016 a kit, HISTO SPOT HLA AB®, which allow to perform both the high resolution searching and identification of anti-HLA antibodies in patient sera. The antibodies profiles obtained with the kit show variable consistency rates with another more used test depending on the sera tested. The goal of the internship was to certify the coherence of the kits results by studying the impact of the quality of the antigens on the kit results. This work should permit Innobiochips to improve the methods used in quality control of the product in order to allow a reliable diagnostic for the patients. Specificly, I disgned an experimental protocol based on the measurement of antibody-antigen affinity which was correlated with clinical results from other technologies. As thoses technologies are high througput ones, I worked with big amounts of data. Therefor, I've learned a lot in data management and statistical analysis. I used Rstudio to perform my analysis and succesfully wrote a program combining Rstudio and LaTeX to analyse in a few seconds a large amount of experimental data and summerise them in a 5 pages pdf document. This internship was a very exciting and rewarding experience.
