Investigated the expression and binding reactivity of highly conserved immunoglobulins (Ig) frequently expressed by chronic lymphocytic leukemia (CLL) B cells. CLL is a lymphoproliferative disease characterized by the expansion of B lymphocytes, many of which express antibodies that have highly similar molecular structures and are polyreactive. Using a recombinant vector system, I co-expressed the Ig heavy and light chains in mammalian cells following transient transfection. Expressed antibodies were detected and quantitated by ELISA, and binding reactivity would be analyzed by ELISA and hybridization to protein arrays containing self-antigens. Show less
